目的:研究细胞周期依赖性激酶2(Cdk2)在NO、内皮素1(ET-1)及血管紧张素Ⅱ(AngⅡ)促血管平滑肌细胞(VSMC)增殖中的作用。方法:应用半定量RT-PCR法测定样品Cdk2水平;3H-TdR参入方法测定细胞DNA合成水平。结果:自发性高血压大鼠(SHR)VSMC中Cdk2的表达明显高于WistarKyotO大鼠(WKY),细胞增殖较快;硝普钠抑制SHRVSMC增殖,CdkZ表达下降;而左旋硝基精氨酸(L-NNA)和ET-1、AugⅡ能够促进WKYVSMCCdk2mRNA的表达和细胞增殖。结论:NO及ET-1、AngⅡ在对VSMC增殖的调节中,Cdk2可能起着重要的作用。
Objective: To investigate the role of Cdk2 in the proliferation of vascular smooth muscle cells (VSMCs) induced by NO, ET-1 and Ang Ⅱ. Methods: The level of Cdk2 was determined by semi-quantitative RT-PCR and 3H-TdR incorporation was used to determine the level of DNA synthesis. Results: The expression of Cdk2 in SHR VSMCs was significantly higher than that in WistarKyotO rats (WKY), and SNP inhibited the proliferation and the expression of CdkZ in SHRVSMC. However, L-NNA L-NNA) and ET-1, AugⅡ can promote WKYVSMCCdk2mRNA expression and cell proliferation. Conclusion: Cdk2 may play an important role in the regulation of VSMC proliferation by NO, ET-1 and AngⅡ.
