目的:探查电针刺激大鼠背部L3-5夹脊穴对大鼠后肢缺血后皮肤血流、温度、肌力以及血清中肌酸激酶(CK)、谷草转氨酶(AST)、乳酸脱氢酶(LDH)含量变化规律。方法:取36只SD大鼠,随机均分为对照组、模型组、A与B实验组,共4组。对照组仅在大鼠双侧骼腰动脉位置柔和地游离腹主动脉及双侧的腹壁阴部动脉。模型组、A、B三组结扎与对照组所暴露位置一致的同名动脉,其中模型组结扎动脉后无治疗处置,A组定时针刺夹脊穴(不通电),B组定时电针夹脊穴。造模成功后于第7天观察大鼠后肢皮肤血流、温度、肌力改变。采用酶联免疫吸附法(ELISA)测定血清中CK、AST、LDH的含量。结果:①大鼠后肢缺血造模成功后出现后肢皮肤血流减慢、皮肤温度降低、肌力下降等改变;②一定时间内,大鼠血清中CK、AST、LDH的含量在后肢缺血造模后增多,与对照组比较差异显著(P<0.05);③电针刺激7d后,大鼠后肢症状体征有所改善,血清CK、AST、LDH含量有所下降,与模型组比较差异显著(P<0.05);④电针组数据变化更加明显,一定程度上优于未通电组。结论:合理电针刺激大鼠L3-5夹脊穴,可使大鼠后肢血管有一定程度舒张作用,减轻机体组织缺血及受损组织血清酶的释放。
Aims: To investigate the effects of electroacupuncture on L3-5 JiaJi acupoint on the skin blood flow, temperature, muscle strength and serum creatine kinase (CK), aspartate aminotransferase (AST), lactate dehydrogenase (LDH) content changes. Methods: 36 SD rats were randomly divided into control group, model group, A and B experimental group, a total of 4 groups. The control group only softly separated the abdominal aorta and bilateral abdominal pubic artery at bilateral iliac arteries in rats. Model group, A, B three groups of the same place of exposure of the same position of the exposed artery of the same name, wherein the model group no treatment after ligation of the artery, A group of acupuncture Jiaji points (no electricity), B group hole. On the 7th day after model establishment, the blood flow, temperature and muscle strength of hind limbs were observed. Serum CK, AST, LDH levels were determined by enzyme-linked immunosorbent assay (ELISA). Results: (1) Hindlimb skin blood flow slowed down, skin temperature decreased and muscle strength decreased after success of rat hindlimb ischemia modeling. (2) The contents of CK, AST and LDH in rat serum were decreased in hindlimb ischemia Compared with the control group, the difference was significant (P <0.05); ③The symptoms and signs of hind limbs were improved after 7 days of electroacupuncture and the levels of serum CK, AST and LDH were decreased, which were significantly different from the model group (P <0.05). (4) The data of electroacupuncture group changed more obviously, which was better than that of non-electrified group. Conclusion: Acupuncture stimulation of L3-5 JiaJi acupuncture can induce a certain degree of vasodilatation in the hindlimb blood vessels of rats and reduce the release of serum enzyme and tissue ischemia.
