目的建立测定肉制品中合成着色剂苋菜红、胭脂红和诱惑红含量的固相萃取-高效液相色谱法。方法以氨水-90%乙醇(1+99,V/V)溶液为提取液,超声提取肉制品样品中的着色剂,经PLS固相萃取柱净化,色谱柱为Dikma Spursil C18(4.6 mm×150 mm,5μm)。以乙腈-0.05 mol/L乙酸铵溶液为流动相进行梯度洗脱色谱分离,流速1.0 ml/min,柱温30℃,进样量10μl,检测波长200~700 nm。结果在0.5~25.0μg/ml,苋菜红、胭脂红和诱惑红的峰面积与其相应浓度呈良好相关性,相关系数r分别为0.999 6、0.999 7和0.999 3,方法加标回收率为94.2%~99.1%。结论本方法准确度高,具有良好的重现性和精密度,能快速对肉制品中3种合成着色剂进行定量检测,满足国家限量标准的要求。
OBJECTIVE To establish a solid phase extraction and high performance liquid chromatography (HPLC) method for the determination of amaranth, carmine and seductive red of synthetic colorants in meat products. Methods Colorants in meat samples were extracted by ultrasonic with 90% ethanol (1 + 99, V / V) solution and purified by PLS column. The chromatographic column was Dikma Spursil C18 (4.6 mm × 150 mm, 5 μm). The mobile phase was eluted with acetonitrile-0.05 mol / L ammonium acetate solution. The flow rate was 1.0 ml / min. The column temperature was 30 ℃. The injection volume was 10μl and the detection wavelength was 200-700 nm. The results showed that there was a good correlation between the peak areas of amaranth, carmine and seductive red at the concentrations of 0.5 ~ 25.0μg / ml with their corresponding concentrations, the correlation coefficients r were 0.999 6,0.999 7 and 0.999 3, respectively. The recoveries were 94.2% ~ 99.1%. Conclusion The method has high accuracy, good reproducibility and precision, and can quickly detect the three kinds of synthetic colorants in meat products to meet the requirements of the national limit standards.
