目的 探讨樟柳碱和山莨菪碱对急性肺损伤(ALI) 的预防作用及其机制。方法 将50 只Wistar 大鼠随机分成5 组:生理盐水组,油酸模型组,地塞米松预防组,樟柳碱预防组,山莨菪碱预防组。各预防组在注入油酸前30 分钟分别于腹腔内注入地塞米松2 mg/kg,樟柳碱2 mg/kg,山莨菪碱10 mg/kg。以上各组均在注油酸后4 小时采血测超氧化物歧化酶(SOD) 、脂质过氧化物(LPO),观察肺组织病理形态学变化。结果 樟柳碱和山莨菪碱预防组血浆LPO含量[(9-09 ±2-07)μmoI/L和(8-67±2-10)μmoI/L] 与油酸模型组血浆LPO含量[(12-18±1-67)μmoI/L] 比较均明显降低( P均< 0-01)。病理组织学观察示樟柳碱和山莨菪碱预防组肺组织病理改变较油酸模型组减轻。结论 樟柳碱、山莨菪碱能改善油酸致ALI的氧自由基及肺的病理形态学变化,对ALI有预防作用。
Objective To investigate the preventive effect of anisodine and anisodamine on acute lung injury (ALI) and its mechanism. Methods Fifty Wistar rats were randomly divided into five groups: saline group, oleic acid model group, dexamethasone prophylaxis group, anisodine prophylaxis group and anisodamine prophylaxis group. Each prophylaxis group was injected with dexamethasone 2 mg / kg, anisodine 2 mg / kg and anisodamine 10 mg / kg intraperitoneally 30 minutes prior to the injection of oleic acid. All the above groups were measured 4 hours after the oleic acid, blood samples were measured for superoxide dismutase (SOD), lipid peroxides (LPO), and histopathological changes were observed. Results In the anisodine and anisodamine preventive groups, the plasma LPO content [(9-09 ± 2-07) μmoI / L and (8-67 ± 2-10) μmoI / L] 12-18 ± 1-67) μmoI / L] (all P <0-01). Histopathological observation showed that anisodamine and anisodamine preventive group lung tissue pathological changes than oleic acid model group alleviate. Conclusion Anisodine and anisodamine can improve the oxygen free radical induced by oleic acid and the pathological changes of lung, which may prevent ALI.
