目的利用扩增片段长度多态性(AFLP)分子标记技术对陕西秦岭地区6个天然居群的三叶木通种质进行遗传多样性分析。方法利用10对引物组合对111份三叶木通种质进行AFLP扩增,并评估遗传多样性。结果 AFLP扩增结果共产生221条清晰的条带,其中多态性条带132条,占59.7%。6个三叶木通居群间遗传分化系数为0.505,基因多态性指数为0.185。在Nei’s遗传距离0.13处,6个天然居群被聚为4个类群。结论陕西秦岭地区三叶木通天然居群遗传多样性水平较低,居群内与居群间的遗传变异程度基本相同。
OBJECTIVE: To analyze the genetic diversity of Cloverleaf through the amplified fragment length polymorphism (AFLP) markers in six natural populations in Qinling Mountains, Shaanxi Province. Methods Ten pairs of primer combinations were used to amplify 111 accessions of Clover. The genetic diversity was evaluated. Results A total of 221 clear bands were produced by AFLP amplification, of which 132 were polymorphic, accounting for 59.7%. The genetic differentiation coefficient of 6 Clover populations was 0.505, and the genetic polymorphism index was 0.185. At Nei’s genetic distance of 0.13, six natural populations were clustered into four groups. Conclusion The genetic diversity of the natural populations of Clover is low in the Qinling Mountains of Shaanxi Province, and the genetic variation within the populations is basically the same as that of the populations.
