目的:研究癌基因HER-2在不同程度宫颈病变脱落细胞的表达,探讨它在宫颈癌发生发展中的作用及临床意义。方法:用荧光原位杂交(FISH)法检测12例正常宫颈、55例宫颈上皮内瘤变(CIN)及24例浸润性宫颈癌脱落细胞中HER-2的表达。结果:HER-2在正常宫颈脱落细胞中不表达,在CINⅠ、CINⅡ~Ⅲ及宫颈癌的阳性表达率分别为6.3%、25.6%及58.3%。HER-2在CINⅡ~Ⅲ及子宫颈癌组表达均显著高于正常组(P<0.01),同时宫颈癌组HER-2明显高于CIN组(P<0.01)。HER-2高表达与细胞分级明显相关(P<0.01),与临床分期、病理类型无关。结论:HER-2基因高表达在宫颈癌发生发展中可能起重要作用,在宫颈病变脱落细胞中检测HER-2基因对探讨宫颈病变的进展和预测不良预后可能有重要意义。
Objective: To study the expression of oncogene HER-2 in exfoliated cells with varying degrees of cervical lesions and its role in the development of cervical cancer and its clinical significance. Methods: The expression of HER-2 in 12 cases of normal cervix, 55 cases of cervical intraepithelial neoplasia (CIN) and 24 cases of invasive cervical cancer exfoliated cells were detected by fluorescence in situ hybridization (FISH). Results: HER-2 was not expressed in normal cervical exfoliated cells. The positive rates of CINⅠ, CINⅡ-Ⅲ and cervical cancer were 6.3%, 25.6% and 58.3% respectively. The expression of HER-2 in CINⅡ-Ⅲ and cervical cancer group was significantly higher than that in normal group (P <0.01), while HER-2 in cervical cancer group was significantly higher than that in CIN group (P <0.01). HER-2 overexpression was significantly associated with cell grade (P <0.01), but not with clinical stage and pathological type. Conclusion: The high expression of HER-2 gene may play an important role in the occurrence and development of cervical cancer. Detecting HER-2 gene in cervical exfoliated cells may play an important role in exploring the progression of cervical lesions and predicting poor prognosis.
