目的:建立测定马比木根中喜树碱和9-甲氧基喜树碱的高效液相色谱法。方法:采用5种方法对马比木根中的喜树碱和9-甲氧基喜树碱进行提取;运用高效液相色谱法,采用Waters Symmetry C18色谱柱(4.6 mm×250 mm,5μm),以甲醇-水(50∶50)为流动相,流速0.8 mL·min-1,检测波长254 nm,柱温26℃。结果:喜树碱和9-甲氧基喜树碱进样量分别在0.24~4.00μg(r=0.9997)和0.24~4.00μg(r=0.9995)范围内线性关系良好;乙醇溶液室温浸泡提取得到的喜树碱含量(0.944%)和9-甲氧基喜树碱含量(0.237%)明显高于其他方法。马比木是目前发现的喜树碱类含量最高的植物。结论:本文建立的HPLC检测法简便、准确、灵敏度高,乙醇溶液室温浸泡提取法安全,成本低,提取率高,适用于喜树碱类提取和马比木的质量检测。
Objective: To establish a HPLC method for determination of camptothecin and 9-methoxycamptothecin in mrubbery root. METHODS: Camptothecin and 9-methoxycamptothecin were extracted from five kinds of moths by using five methods. The HPLC-MS was performed on a Waters Symmetry C18 column (4.6 mm × 250 mm, 5 μm) The mobile phase was methanol-water (50:50) with a flow rate of 0.8 mL · min-1. The detection wavelength was 254 nm and the column temperature was 26 ℃. Results: The injection volume of camptothecin and 9-methoxycamptothecin was linear in the range of 0.24-4.00μg (r = 0.9997) and 0.24-4.00μg (r = 0.9995) respectively. The ethanol solution was soaked and extracted at room temperature Of the camptothecin content (0.944%) and 9-methoxy camptothecin content (0.237%) were significantly higher than other methods. Mabi is the most camptothecin-rich plant ever discovered. Conclusion: The HPLC method established in this paper is simple, accurate and sensitive. The room temperature immersion extraction method of ethanol solution is safe, low cost and high extraction rate. It is suitable for the extraction of camptothecins and the quality control of Machilus wood.
