本项研究取得了一种水稻原生质体高效再生为可育植株的方法。以水稻栽培品种Lemont,Laccasin,Cypress,Mercury,Lebelle,89-2,秀水04,135-3542,台北309和Nibbonbare的幼穗为材料诱导愈伤组织,以改良型R2培养基进行悬浮培养。在三个供试培养基中,R2培养基最适合胚状体的发生。悬浮细胞系游离获得的原生质体产量很高,可达每克鲜重106~3×107个。所有供试水稻品种都获得了再生植株。
In this study, we obtained a method for efficient regeneration of rice protoplasts into fertile plants. The calli were induced from the young ear of rice cultivars Lemont, Laccasin, Cypress, Mercury, Lebelle, 89-2, Xiushui 04, 135-3542, Taipei 309 and Nibbonbare, and cultured in modified R2 medium. In three test media, R2 medium best suited for embryoid body. The free protoplasts obtained from suspension cell lines have very high yield, reaching 106 to 3 × 107 per gram of fresh weight. All tested rice varieties were regenerated.
