目的:研究降糖消渴颗粒含药血清对胰岛瘤细胞INS-1凋亡的影响并对其作用机制进行初步探讨。方法:应用中药血清药理学方法制备降糖消渴颗粒含药血清及空白血清,以大鼠胰岛瘤细胞INS-1为研究对象,观察降糖消渴颗粒含药血清对INS-1细胞形态的影响,并用流式细胞术检测含药血清对细胞凋亡率的影响,用Western blot的方法检测Bax和Bcl-2蛋白表达水平,初步探讨其抗凋亡机制。结果:与空白血清组比较,降糖消渴颗粒含药血清可保护INS-1细胞形态,并降低细胞凋亡率,同时降糖消渴颗粒含药血清减少了Bax蛋白表达,增加Bcl-2蛋白表达,提高了Bcl-2/Bax蛋白的比值。结论:降糖消渴颗粒含药血清可抑制胰岛瘤细胞的INS-1凋亡,其机制可能与以线粒体为核心的凋亡途径有关。
Objective: To study the effect of Jiangtangxiaoke granule-containing serum on the apoptosis of INS-1 cells and to explore its mechanism. Methods: The Chinese medicine serum pharmacology method was used to prepare the drug-containing serum of Jiangtangxiaoke granule and the blank serum. INS-1 cells of rat’s insulinoma cells were used as research object to observe the effects of Jiangshe Xiaoke granules containing serum on INS-1 cell morphology The effect of containing serum on the apoptosis rate was detected by flow cytometry. The protein expression of Bax and Bcl-2 was detected by Western blot, and the anti-apoptotic mechanism was preliminarily discussed. Results: Compared with the blank serum group, the drug-containing serum of Jiangtangxiaomai Granule could protect the morphology of INS-1 cells and reduce the apoptosis rate. At the same time, the drug-containing serum of Jiangtangxiemai granule reduced Bax protein expression and increased Bcl-2 Protein expression, increased Bcl-2 / Bax protein ratio. CONCLUSION: Jiangtang Xiaoke granule-containing serum can inhibit INS-1 apoptosis of islet cells, which may be related to the mitochondria-mediated apoptotic pathway.
