目的:建立环香酮滴眼液中秦皮甲素含量测定方法。方法:HPLC采用YMC C18柱(250 mm×4.6 mm,10μm),乙腈-0.1%磷酸(12∶88)为流动相,流速为1.0 mL.min-1,检测波长为334 nm,柱温为30℃。结果:秦皮甲素在0.028 4~0.284μg范围内线性关系良好(r=0.999 6),平均加样回收率为99.16%,RSD为0.89%;测得每瓶含秦皮甲素大于1.51 mg。结论:该方法重现性好,准确度高,可作为环香酮滴眼液定量分析方法。
Objective: To establish a method for determination of aesculin in eyedrops of chinone. METHODS: HPLC was performed on a YMC C18 column (250 mm×4.6 mm, 10 μm) with acetonitrile-0.1% phosphoric acid (12:88) as the mobile phase, a flow rate of 1.0 mL.min-1, a detection wavelength of 334 nm, and a column temperature of 30. °C. RESULTS: There was good linearity in the range of 0.028 4~0.284 μg (r=0.999 6), the average recovery rate was 99.16%, RSD was 0.89%, and each bottle contained more than 1.51 mg of aesculin. Conclusion: This method has good reproducibility and high accuracy. It can be used as a quantitative analysis method for chinone ketone eye drops.
