随着抗性细胞突变体筛选的研究的深入,继而产生的突出问题是随着继代培养时间的延长,离体培养组织和细胞的分化及再生植株的能力逐渐下降以至完全消失。关于如何提高高代愈伤组织分化频率,国内外已有许多报道,主要涉及后代染色体的变化,至于激素对酶的影响在高代中尚未见到,为此,我们以高代水稻花药愈伤组织(继代15次培养600天直至继代19次培养天数720天)为试材,研究了不同激素、琼脂、甘露醇对高代愈伤组织分化的影响,以及在分化过程中呼吸强度和同工酶的变化。
