外周血CD_(34)抗原阳性,CD_(33)抗原阴性的细胞,在造血细胞生长因子(HGF)──G-CSF,IL-3和干细胞因子(SCF)的联合作用下,可以在体外维持造血达9周,克隆形成细胞(CFC)大量增加。CFC和有核细胞增殖高峰在液体培养的第6~7周。在液体培养中不加入HGF和单独应用G-CSF的情况下,这类细胞不能在体外增殖。结果表明,HGF的联合应用在维持外周血造血细胞的体外长期生长起主要作用。
Peripheral CD34 (34) antigen-negative and CD33 (33) -negative cells can be maintained in vitro under the combined action of hematopoietic growth factor (HGF) ──G-CSF, IL-3 and stem cell factor (SCF) After 9 weeks of hematopoiesis, the number of clonogenic cells (CFCs) increased greatly. The peak of CFC and nucleated cell proliferation was in the sixth to seventh week of liquid culture. In the absence of HGF and G-CSF alone in liquid culture, such cells do not proliferate in vitro. The results indicate that the combined use of HGF plays a major role in maintaining the long-term growth of peripheral blood hematopoietic cells in vitro.
