目的观察岷当归有效成分当归挥发油、当归多糖、阿魏酸对高同型半胱氨酸血症(HHcy)兔腹主动脉血管弹性的影响。方法取日本大耳白兔48只,按随机数字表法分为6组,即正常组、模型组、叶酸维生素组(叶酸1.5 mg·kg~(-1)·d~(-1)+维生素B_6 20 mg·kg~(-1)·d~(-1)+维生素B_(12) 0.25mg·kg~(-1)·d~(-1))、当归挥发油组(100 mg·kg~(-1)·d~(-1))、当归多糖组(120 mg·kg~(-1)·d~(-1))和阿魏酸组(2.5 mg·kg~(-1)·d~(-1)),每组8只。采用DL-蛋氨酸皮下注射法建立HHcy家兔模型。造模8周后各给药组给予相应药物灌胃。12周后检测各组同型半胱氨酸(Hcy)水平,观察腹主动脉血管病理变化,行腹主动脉血管超声检查,测量腹主动脉内-中膜厚度(IMT)、腹主动脉血流峰值(Vp)、舒张末期内径(D_1)和收缩末期内径(D_2),计算血管弹性(D_1-D_2)。结果模型组血清Hcy水平高于正常组(P<0.05);与模型组比较,当归挥发油组、阿魏酸组、叶酸维生素组血清Hcy水平明显降低(P<0.05)。与正常组比较,模型组IMT和Vp增加(P<0.05),动脉弹性值减小(P<0.05);与模型组比较,叶酸维生素组、当归挥发油组、当归多糖组、阿魏酸组IMT、Vp值减少(P<0.05),动脉弹性值增加(P<0.05)。腹主动脉病理学检查发现模型组有明显的AS征象,当归挥发油组、阿魏酸组病变程度较模型组减轻。结论岷当归有效成分当归挥发油、阿魏酸能够降低血清Hcy水平,减轻血管硬化病变,对HHcy所致的AS病变具有一定的防治作用。
Objective To observe the effects of Angelica sinensis (Angelica sinensis) Angelica Sinensis, Angelica sinensis polysaccharide and ferulic acid on the vascular elasticity of abdominal aorta in hyperhomocysteinemia (HHcy) rabbits. Methods Forty-eight Japanese white rabbits were divided into 6 groups according to random number table: normal group, model group, folic acid and vitamin group (folic acid 1.5 mg · kg -1 · d -1) B_6 20 mg · kg -1 · d -1 · Vitamin B 12.25 mg · kg -1 · d -1), and Angelica sinensis volatile oil group (100 mg · kg -1 · d -1) (-1) · d ~ (-1)), 120 mg · kg -1 · d -1 and 2.5 mg · kg -1 · d ~ (-1)), 8 in each group. HHcy rabbit model was established by subcutaneous injection of DL-methionine. After 8 weeks of modeling, each administration group was given the corresponding drug gavage. After 12 weeks, the levels of homocysteine (Hcy) in each group were detected, the pathological changes of abdominal aorta were observed, and abdominal aortic vascular ultrasound examination was performed to measure the aortic intima-media thickness (IMT), abdominal aorta blood flow (Vp), end-diastolic diameter (D_1) and systolic end-diastolic diameter (D_2) were measured to calculate the blood vessel elasticity (D_1 -D_2). Results Compared with the model group, the Hcy level in the model group was significantly higher than that in the normal group (P <0.05). Compared with the model group, the Hcy level in the Angelica sinensis volatile oil group, the ferulic acid group and the folic acid vitamin group was significantly lower (P <0.05). Compared with the normal group, the IMT and Vp in the model group increased (P <0.05) and the arterial elasticity decreased (P <0.05). Compared with the model group, the folic acid and vitamin D group, Angelica sinensis polysaccharide group, , Vp decreased (P <0.05) and arterial elasticity increased (P <0.05). Abdominal aorta pathological examination found that the model group had obvious signs of AS, Angelica volatile oil group, ferulic acid group lesion than the model group. Conclusion Angelica sinensis Angelica sinensis active ingredient volatile oil, ferulic acid can reduce serum Hcy levels, reduce vascular sclerosis, HHcy-induced AS lesions have a preventive effect.
