目的检测环氧化酶-2(COX-2)、血管内皮生长因子(VEGF)和CD105在人膀胱移行细胞癌(TCCB)组织和正常膀胱组织中的表达,探讨COX-2和VEGF的表达与血管形成的关系。方法采用RT-PCR法检测人TCCB和正常膀胱组织中COX-2 mRNA的表达。采用免疫组织化学法检测人TCCB和正常膀胱组织中VEGF和CD105的表达并对微血管进行计数。结果正常膀胱组织不表达COX-2 mRNA,VEGF和CD105的表达很低;人TCCB组织中COX-2 mRNA、VEGF和CD105的表达均显著高于正常膀胱组织(P<0.01或<0.05)。低分化和肌层浸润型癌组织中COX-2 mRNA、VEGF和CD105表达显著高于高分化和非肌层浸润型癌组织(P<0.01或<0.05)。结论COX-2和VEGF与人TCCB的分级和分期有关。COX-2和VEGF与癌组织中血管形成密切相关。
Objective To investigate the expression of cyclooxygenase-2 (COX-2), vascular endothelial growth factor (VEGF) and CD105 in human bladder transitional cell carcinoma (TCCB) and normal bladder tissues, The relationship between the formation of blood vessels. Methods RT-PCR was used to detect the expression of COX-2 mRNA in human TCCB and normal bladder tissues. Immunohistochemistry was used to detect the expression of VEGF and CD105 in human TCCB and normal bladder tissues and the microvessels were counted. Results The expression of COX-2 mRNA, VEGF and CD105 in normal bladder tissue was lower than that in normal bladder tissue (P <0.01 or <0.05). The expression of COX-2 mRNA, VEGF and CD105 in human TCCB tissues was significantly higher than that in normal bladder tissues. The expression of COX-2 mRNA, VEGF and CD105 in poorly differentiated and myometrial invasive carcinoma was significantly higher than that in well differentiated and non-myometrial invasive carcinoma (P <0.01 or <0.05). Conclusions COX-2 and VEGF are related to the classification and staging of human TCCB. COX-2 and VEGF are closely related to the formation of blood vessels in cancer tissues.
