目的探讨新型免疫分子TIPE2对肝癌细胞转移与侵袭的影响作用。方法取35例肝癌患者外周血单核细胞及40例健康对照样本,采用实时荧光定量法检测外周血PBMCs中TIPE2的表达水平,采用划痕实验和Transwell试验检测TIPE2对BEL7402细胞转移与侵袭的影响。结果肝癌病人PBMCs中TIPE2的表达下调,且其下调程度与肿瘤分期相关,细胞转染实验表明TIPE2上调可以抑制肝癌细胞系的转移与侵袭。结论 TIPE2可能作为一种新型的抑癌基因在肿瘤的发生发展中起重要生物学作用。
Objective To investigate the effect of TIPE2, a novel immune molecule, on the metastasis and invasion of hepatocellular carcinoma cells. Methods Peripheral blood mononuclear cells from 35 HCC patients and 40 healthy controls were collected. The expression of TIPE2 in peripheral blood PBMCs was detected by real-time fluorescence quantitative assay. The scratch and Transwell assays were used to detect the effect of TIPE2 on the metastasis and invasion of BEL7402 cells . Results The expression of TIPE2 was down-regulated in PBMCs from patients with hepatocellular carcinoma. The down-regulation of TIPE2 was correlated with tumor stage. The transfection experiments showed that up-regulation of TIPE2 could inhibit the metastasis and invasion of hepatocellular carcinoma cell lines. Conclusion TIPE2 may play an important biological role as a novel tumor suppressor gene in tumorigenesis.
