目的 探讨靶向抑制CXCR7基因表达对人膀胱癌RT4细胞增殖的影响,初步分析其作用机制.方法 采用小干扰RNA(Small interfering RNA,siRNA)干扰技术沉默人膀胱癌RT4细胞中CXCR7基因的表达,分别采用qRT-PCR和Western blot检测siRNA的靶向沉默效果.通过CCK-8增殖实验研究CXCR7抑制对RT4细胞增殖的影响.通过Western blot检测RT4中Akt通路关键因子Akt、p-Akt、Bad及pBad的表达,初步探讨CXCR7抑制调控RT4细胞增殖的分子机制.结果 qRT-PCR和Western blot检测结果显示,转染CXCR7 siRNA质粒可显著抑制RT4细胞中CXCR7mRNA和蛋白表达.CXCR7干扰组细胞生长速度被显著抑制(P<0.05),CXCR7干扰组细胞中pAkt及pBad表达水平较对照组显著减少(P<0.05).结论 siRNA干扰靶向抑制CXCR7表达抑制RT4细胞的增殖,可能与其抑制Akt信号通路有关.“,”Objective To study the effect and mechanism of CXCR7 gene silence by siRNA on the proliferation ability of human bladder cancer cell line RT4.Methods CXCR7 siRNA was transfected into bladder cancer RT4 cells.48h later,qRT-PCR and Western blot were used to detect the expression of CXCR7 in RT4 cells to study the silencing effect,CCK8 assay was performed to evaluate the proliferation of RT4 cells,and Western blot was used to detect the expressions of Akt,pAkt,Bad and pBad.Results The expression levels of CXCR7 mRNA and protein were decreased,the proliferation rate of RT4 cells and the expression levels of pAkt and pBad in RT4 cells were all obviously decreased in the transfected group than in negative control group or the blank control group (P < 0.05).Conclusion CXCR7 siRNA inhibited the proliferation of RT4 cells,which may be due to its inhibitive effect on Akt pathway.
