内毒素脂多糖是革兰阴性菌死亡解体后释放的致病因子,严重威胁人类健康。膜转运体与药物/毒物的药动学性质、体内的安全有效具有密切关系,而肠道、肝脏、肾脏及脑组织是体内转运体表达的主要场所。近年来研究发现,内毒素脂多糖会引起转运体的表达及功能的变化,进而引起药物/毒性物质在相关组织暴露变化,甚至引起肠道屏障损伤、肝肾毒性及中枢系统疾病的发生。因此研究内毒素脂多糖对药物转运体的影响具有极其重要的意义。本文针对内毒素脂多糖对药物转运体的影响展开调研,并对其对肠道、肝脏、肾脏及脑等器官转运体表达及功能的影响研究进展进行文献综述。
Endotoxin lipopolysaccharide is a causal agent of Gram-negative bacteria released after death and seriously threatens human health. Membrane transporters are closely related to the pharmacokinetic properties of drugs / poisons and safe and effective in vivo. Intestinal, liver, kidney and brain tissues are the main sites for the expression of transporters in vivo. In recent years, studies have found that lipopolysaccharide endotoxin can cause changes in the expression of transporter and function, and then lead to drug / toxic substances in the relevant tissue changes in exposure, and even cause intestinal barrier injury, liver and kidney toxicity and central system diseases. Therefore, the study of lipopolysaccharide endotoxin on the impact of drug transporter has an extremely important significance. In this paper, we investigated the effects of lipopolysaccharide (LPS) on drug transporters and reviewed the research progress on the effects of LPS on the expression and function of intestinal transporters such as gut, liver, kidney and brain.
