应用高压液相色谱法测定人体血浆内阿米替林及其主要代谢产物去甲替林及羟基去甲替林的血药浓度。以产国硅胶YQG—80(5μm)为固定相,甲醇—水极性流动相,三甲丙咪嗪及普罗替林为双内标物。紫外检测波长240nm。阿米替林、去甲替林和羟基去甲替林的最低检出限量分别为2,5和5ng,最低检出浓度分别为4,10和10ng/ml(血浆),回收率分别为>94,97和97%。方法简便,灵敏度高,适于临床药理研究及临床治疗监测。
Determination of plasma concentrations of amitriptyline and its major metabolites, nortriotine and hydroxybenbuterol, in human plasma by high pressure liquid chromatography. Chromatography was performed on a YQG-80 (5 μm) -oriented silica gel with methanol-water polar mobile phase, trimeprazide and protriptyline as double internal standards. UV detection wavelength of 240nm. The minimum detectable levels of amitriptyline, nortriptyline and hydroxy nortriptyline were 2, 5 and 5 ng, respectively, with the lowest detectable concentrations of 4, 10 and 10 ng / ml (plasma), respectively. The recovery rates were> 94, 97 and 97%. The method is simple, high sensitivity, suitable for clinical pharmacology research and clinical treatment monitoring.
