目的:建立小鼠免疫性肝损伤模型,研究剑叶耳草水提物对小鼠免疫性肝损伤的保护作用。方法:采用刀豆蛋白A(Con A)尾静脉注射建立小鼠免疫性肝损伤模型,用比色分析法测定小鼠血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、碱性磷酸酶(ALP)、总胆红素(TBIL)的活性;测定小鼠肝匀浆丙二醛(MDA)、还原型谷胱甘肽(GSH)的含量及谷胱甘肽过氧化物酶(GSH-Px)、过氧化物歧化酶(SOD)的活性;HE染色观察各组小鼠肝细胞损伤程度。结果:8g/kg和4g/kg剑叶耳草水提物能显著降低因Con A所致免疫性肝损伤小鼠血清异常升高的ALT、AST、ALP和TBIL;对免疫性肝损伤小鼠肝匀浆中MDA的含量明显降低,GSH含量明显升高及GSH-Px、SOD的活性有明显的升高作用。结论:剑叶耳草水提物对ConA诱导的免疫性肝损伤小鼠具有一定的保护作用,其机制可能与其抗氧自由基损伤、抑制脂质过氧化作用有关。
OBJECTIVE: To establish a mouse model of immunological liver injury and to study the protective effect of Aqueous extract of Scutellaria barbata on immunological liver injury in mice. Methods: A mouse model of autoimmune liver injury was established by tail vein injection of concanavalin A (Con A) and the levels of serum ALT, AST and ALP (aspartate aminotransferase (ALP) and total bilirubin (TBIL). The contents of malondialdehyde (MDA), reduced glutathione (GSH) and glutathione peroxidase (GSH-Px ), Superoxide dismutase (SOD) activity; HE staining observed the degree of liver cell injury in each group. Results: The aqueous extracts of 8g / kg and 4g / kg Acridothera indica could significantly reduce the levels of ALT, AST, ALP and TBIL in mice with immunological liver injury induced by Con A, Liver homogenate MDA content was significantly reduced, GSH content was significantly increased and GSH-Px, SOD activity was significantly increased. CONCLUSION: The aqueous extract of Scutellaria barbata has a protective effect on ConA-induced immune liver injury in mice. The mechanism may be related to its antioxidant free radical damage and inhibition of lipid peroxidation.
