将巴西固氮螺菌(Azospirillumbrasilense)nifH-lacZ转入粪产碱菌(Alcaligenesfaecalis)A1501中,β-Galactosidast3活性测定结果大明.该转录型融合基因nifH-lacZ在A1501中的表达为诱导型的.在固氮条件下高水平表达,在高铵好氧下低水平表达,在无铵好氧下部分表达。Southern杂交分析证明在粪产碱菌中存在nifA、rpoN和ntrC基因的同源序列,表明粪产碱菌nifHDK的转录启始可能为NifA和α依赖型的。携带nifH-lacZ融合基因的粪产碱菌结合子在宿主水稻根表定位(接种18h)后.nifH-lacZ开始表达.结合子定殖(接种24h)后,开始表现出乙炔还原活性,接种72h后,nifH-lacZ表达值和固氮活性达到最大值。
The Azifirillumbrasilense nifH-lacZ was transferred into Alcaligenes faecalis A1501, and the result of β-Galactosidast3 activity was determined. Expression of the transcriptional fusion gene nifH-lacZ in A1501 was inducible. It is highly expressed under nitrogen fixation, expressed at low levels under high ammonium and aerobic, and partially expressed under ammonium-free aerobic conditions. Southern hybridization analysis demonstrated the presence of homologous sequences of the nifA, rpoN and ntrC genes in Alcaligenes faecalis, suggesting that initiation of nifHDK transcription may be NifA and alpha-dependent. The Alcaligenes faecalis binders carrying the nifH-lacZ fusion gene were located on the root surface of the host rice (18 h after inoculation). nifH-lacZ began to express. After 24 hours of inoculation, the acetylene reduction activity began to show. After 72 hours of inoculation, the expression value of nifH-lacZ and the activity of nitrogen fixation reached the maximum.
