目的研究肺腺癌转移相关转录子1(MALAT1)对人宫颈癌癌Hela细胞顺铂化疗敏感性的影响。方法用RNA干扰(RNAi)技术下调MALAT1在人宫颈癌Hela细胞中的表达,对Hela细胞用顺铂1 mg·L-1进行处理。用细胞计数法检测不同时间点Hela细胞增殖能力的变化,用流式细胞检测Hela细胞周期变化。结果 RNAi下调MALAT1表达后,Hela细胞对顺铂化疗敏感性显著增高,而细胞增殖能力明显下降(均P<0.05)。G0/G1期细胞比率明显增高(P<0.05),Hela细胞迁移能力显著降低(P<0.05)。结论抑制长链非编码RNA MALAT1可增强人宫颈癌Hela细胞对顺铂化疗的敏感性。
Objective To investigate the effect of lung metastasis-associated transcript 1 (MALAT1) on chemosensitivity to cisplatin in human cervical cancer Hela cells. Methods The expression of MALAT1 in human cervical cancer Hela cells was down-regulated by RNA interference (RNAi), and Hela cells were treated with cisplatin 1 mg · L-1. The proliferation of Hela cells at different time points was detected by the cell counting method. The changes of Hela cell cycle were detected by flow cytometry. Results After RNAi down-regulated the expression of MALAT1, the sensitivity of Hela cells to cisplatin chemotherapy was significantly increased, while the cell proliferation ability was significantly decreased (all P <0.05). The ratio of G0 / G1 phase cells was significantly increased (P <0.05), and the migration ability of Hela cells was significantly decreased (P <0.05). Conclusion Inhibition of long-chain non-coding RNA MALAT1 can enhance the sensitivity of human cervical cancer Hela cells to cisplatin chemotherapy.
