目的 研究由玉足海参提取的糖胺聚糖 (GAG)抗血栓形成作用的机制。方法 分别用1mg/L、5mg/L、10mg/L的GAG和 5mg/L肝素与经 1mg/L细菌脂多糖 (LPS)诱导的人脐静脉内皮细胞孵育 ,6h后观察内皮细胞的促凝活性、组织因子 (TF)和凝血酶调节蛋白 (TM)抗原的表达及TF和TMmRNA转录的变化。结果 GAG降低受刺激内皮细胞的促凝活性、TF抗原的表达和TFmRNA的转录 ,升高TM抗原的表达和TMmRNA的转录。结论 下调受刺激内皮细胞TF表达的同时上调TM表达可能是GAG发挥抗血栓形成作用的主要机制之一。
Aim To study the antithrombotic mechanism of glycosaminoglycan (GAG) extracted from jade sea cucumber. Methods Human umbilical vein endothelial cells (HUVECs) were induced by 1 mg / L, 5 mg / L, 10 mg / L GAG and 5 mg / L heparin respectively. The procoagulant activity of endothelial cells was observed after 6 h , Tissue factor (TF) and thrombin regulatory protein (TM) antigen expression and TF and TM mRNA transcription changes. Results GAG decreased pro-coagulant activity of stimulated endothelial cells, TF antigen expression and TF mRNA transcription, increased TM antigen expression and TM mRNA transcription. Conclusion Down-regulation of TF expression by stimulated endothelial cells and up-regulation of TM expression may be one of the main mechanisms by which GAG exerts antithrombotic effect.
