目的:建立人血浆中紫杉醇的高效液相色谱测定法。方法:血浆中紫杉醇用醋酸乙酯提取后在C18柱上以甲醇-水(6535,v/v)为流动相,检测波长227nm,以峰面积外标法定量。结果:该法萃取回收率为94.58%,方法回收率为99.19%;日内、日间RSD均小于4%(n=5),应用本法测定了两例癌症患者静脉输注紫杉醇的经时血药浓度符合二室开放模型。结论:本法操作简便,灵敏度高,专属性强。
Objective: To establish a method for the determination of paclitaxel in human plasma by high performance liquid chromatography. Methods: The plasma paclitaxel was extracted with ethyl acetate on a C18 column with methanol-water (6535, v / v) as the mobile phase at a detection wavelength of 227 nm and quantified by peak area external standard method. Results: The recovery rate of this method was 94.58%, and the recovery rate was 99.19%. The intra-day and inter-day RSDs were less than 4% (n = 5). Two of the cancer patients received intravenous infusion of paclitaxel The time-dependent plasma concentration conforms to the two-compartment open model. Conclusion: This method is easy to operate, high sensitivity and specificity.
