目的 :分别检测宫颈分泌物、血液和宫颈病变组织中的基质金属蛋白酶-9(MMP-9)和转化生长因子β1(TGF-βl)的表达情况,探讨其在宫颈病变形成及演进过程中的意义。方法 :应用ELISA法检测66例病人(其中慢性宫颈炎组织10例、CINcervical intraepithelial neoplasia宫颈上皮内瘤变24例,宫颈鳞癌32例)的宫颈分泌物和血清中的TGF-βl和MMP-9的表达水平。应用免疫组织化学SP法检测相对应病人石蜡包埋的不同病变宫颈组织中TGF-βl和MMP-9的表达。结果 :1.在血清和宫颈分泌物中TGF-βl和MMP-9的浓度与宫颈癌密切相关:慢性宫颈炎组、宫颈上皮内瘤变(CIN)组和宫颈鳞癌(Cx Ca)组的样本中TGF-βl和MMP-9的浓度逐级升高,提示随着宫颈癌恶性程度的增加,TGF-βl和MMP-9的表达明显增强。血清和宫颈分泌物中MMP-9和TGF-β1与其切片免疫组化结果相吻合。随着MMP-9在宫颈分泌物和血清中表达的增强,TGF-β1的表达也明显增高,两者呈正相关。2.TGF-βl和MMP-9表达与宫颈癌的病理和临床分类的关系:在宫颈癌组织中有淋巴结转移的MMP-9和TGFβ1的表达阳性率显著高于无淋巴结转移的。在宫颈癌≥Ⅱb期的MMP-9和TGF-βl的表达阳性率显著高于≤Ⅱa期的。MMP-9和TGF-βl与宫颈癌肿瘤的大小和组织病理学分级无显著性差异。结论 :TGF-βl和MMP-9在宫颈病变、宫颈分泌物和血清的中表达与病变良恶性有关,TGF-βl和MMP-9高表达是宫颈恶性肿瘤的表现之一,并且TGF-βl和MMP-9在宫颈分泌物或血清中的表达具有显著相关性。
OBJECTIVE: To detect the expression of matrix metalloproteinase-9 (MMP-9) and transforming growth factor-β1 (TGF-β1) in cervical secretions, blood and cervical lesions, and to explore the role of MMP-9 in the pathogenesis and progression of cervical lesions significance. Methods: The secretions and serum levels of TGF-β1 and MMP-9 in 66 patients (10 chronic cervicitis tissue, 24 cervical intraepithelial neoplasia cervical intraepithelial neoplasia and 32 cervical squamous cell carcinoma) were detected by ELISA. The level of expression. Immunohistochemical SP method was used to detect the expression of TGF-β1 and MMP-9 in different pathological changes of cervical tissue corresponding to paraffin-embedded patients. The levels of TGF-β1 and MMP-9 in serum and cervical secretions were closely related to cervical cancer: chronic cervicitis group, cervical intraepithelial neoplasia (CIN) group and cervical squamous cell carcinoma (Cx Ca) group The levels of TGF-β1 and MMP-9 in the samples gradually increased, suggesting that with the increase of the malignant degree of cervical cancer, the expressions of TGF-β1 and MMP-9 were significantly increased. Serum and cervical secretions MMP-9 and TGF-β1 consistent with the results of its section immunohistochemistry. With the increased expression of MMP-9 in cervical secretions and serum, the expression of TGF-β1 was also significantly increased, the two were positively correlated. The relationship between the expression of TGF-β1 and MMP-9 and the pathological and clinical classification of cervical cancer: The positive rate of MMP-9 and TGFβ1 with lymph node metastasis in cervical cancer tissues was significantly higher than that without lymph node metastasis. The positive rate of MMP-9 and TGF-β1 expression in stage Ⅱb cervical cancer was significantly higher than that in stage Ⅱa. MMP-9 and TGF-βl and cervical cancer size and histopathological grade no significant difference. Conclusion: The expressions of TGF-β1 and MMP-9 in cervical lesions, cervical secretions and serum are related to benign and malignant lesions. The high expressions of TGF-β1 and MMP-9 are one of the malignant tumors of the cervix. The expression of MMP-9 in cervical secretions or serum had a significant correlation.
