目的研究骨髓间充质干细胞(MSCs)移植对大鼠肺气肿的疗效。方法 Wistar大鼠60只,随机分为对照组、模型组和移植组,每组20只。模型组和移植组大鼠于气管内注入脂多糖并烟雾暴露,建立肺气肿大鼠模型。然后将培养的MSCs经尾静脉注入到移植组大鼠体内,观察移植后3组大鼠肺病理学改变情况。结果模型组和移植组均出现肺气肿病理改变,但后者较前者明显减轻。3组间平均肺泡数(MAN)、肺泡腔与肺总面积比(PAA)差异均有统计学意义(P﹤0.001);移植组MAN高于模型组,但低于正常对照组;PAA低于模型组,但高于正常对照组(P均﹤0.05)。移植组可见5-溴氮尿嘧啶核苷(Brdu)阳性细胞表达,部分Brdu阳性细胞内有肌酸激酶(CK)的表达。对照组和模型组未见Brdu阳性及CK阳性细胞。结论 MSCs移植明显减轻肺气肿大鼠肺部炎症和病理改变,这可能与MSCs在受体肺组织内分化为肺泡上皮细胞有关。
Objective To study the effects of bone marrow mesenchymal stem cells (MSCs) transplantation on emphysema in rats. Methods Sixty Wistar rats were randomly divided into control group, model group and transplantation group, with 20 rats in each group. Rats in model group and transplantation group were implanted with lipopolysaccharide and exposed to smoke in the trachea to establish a rat model of emphysema. Then the cultured MSCs were injected into the tail vein to the transplantation group to observe the pathological changes of the lungs in the 3 groups. Results The pathological changes of emphysema occurred in the model group and the transplantation group, but the latter were significantly less than the former. The mean alveolar number (MAN), total alveolar space and total lung area (PAA) in the three groups were significantly different (P <0.001); MAN in the transplantation group was higher than that in the model group but lower than that in the normal control group Model group, but higher than the normal control group (all P <0.05). In the transplantation group, the expression of Brdu positive cells and the expression of creatine kinase (CK) in some Brdu positive cells were observed. Brdu positive and CK positive cells were not found in the control group and model group. Conclusion MSCs transplantation can significantly reduce lung inflammation and pathological changes in rats with emphysema, which may be related to the differentiation of MSCs into alveolar epithelial cells in the recipient lung tissue.
