肾茶水提液依次用石油醚、氯仿、乙酸乙酯萃取,与水层得到4个不同极性部位。以清除DPPH·、超氧阴离子和羟自由基的能力,络合Fe2+能力,体外抗活性氧能力以及对FeSO4和H2O2诱导小鼠肾脏组织匀浆脂质过氧化产物MDA和GSH-Px水平的影响,综合评价其抗氧化活性;以抑制Fe2+-L-Cys诱导肾脏线粒体肿胀能力,初步评价其保护肾脏线粒体的作用。结果显示肾茶水提液4个不同极性部位均具有不同程度的清除DPPH·、O-·2、·OH和络合Fe2+能力;其中乙酸乙酯和水提部位具有显著体外抗ROS能力,降低MDA和升高GSHPx水平,以及抑制肾脏线粒体肿胀的能力,且其作用效果均与剂量呈正相关。本实验结果可为肾茶用于治疗肾脏疾病的药效物质基础研究提供依据。
The extract of kidney tea extract was extracted with petroleum ether, chloroform and ethyl acetate successively, and obtained 4 different polar sites with water layer. The results showed that the extracts of Radix et Rhizoma Anemarrhenae were able to scavenge DPPH ·, O- · 2 · · OH and complex Fe2 + in four different polarity fractions. Ethyl acetate and water-extractable fractions had significant anti-ROS abilities in vitro and decreased MDA and elevated GSHPx levels, as well as the ability to inhibit the swelling of the kidneys mitochondria, and its effects are positively correlated with the dose. The results of this experiment can provide the basis for the basic research of pharmacodynamics of kidney tea for treating kidney disease.
