自Titzpatrick于1972年从兔主动脉分离出微粒体膜部分(MIC)以来,此分离技术已广泛应用于生化、生理及药理等学科的研究。本实验室在1979年首先采用改进的方法,在大鼠肠系膜动脉组织分离出不同部分的亚细胞膜2。此法虽亦用于大鼠肠系膜静脉,但是由大鼠静脉组织取得的亚细胞膜含量有限,所以本研究利用此改进方法从狗肠系膜静脉获取更多的细胞膜。本文着重阐述采用狗肠系膜静脉制备各亚细胞膜部分的方法,并通过测定相应的标记酶活性,鉴定其纯度。
Since Titzpatrick isolated the microsomal membrane fraction (MIC) from rabbit aorta in 1972, this separation technique has been widely used in biochemical, physiological and pharmacological studies. In 1979, our laboratory first adopted an improved method to separate different subcellular membranes 2 in the rat mesenteric artery. Although this method is also used in rat mesenteric vein, but obtained from the rat venous tissue subcellular membrane content is limited, so this study using this improved method to obtain more membrane from the dog mesenteric vein. This article focuses on the use of dog mesenteric vein preparation of subcellular membrane part of the method, and by measuring the corresponding marker enzyme activity, identification of its purity.
