在建立人主动脉平滑肌细胞(SMC)体外培养方法的基础上,观察了各种天然和氧化修饰型脂蛋白对培养人SMC形态以及表型的影响。在氧化修饰低密度脂蛋白(OX-LDL),氧化修饰高密度脂蛋白(Ox-HDL)和氧化修饰极低密度脂蛋白(OX-VLDL)的作用下,SMC形态发生以下变化:胞体变大,由梭形变为不规则多边形,突起细而长,而且“谷”、“峰”样生长方式也发生改变;LDL和vLDL对SMC的形态亦有影响,但不如氧化修饰脂蛋白明显;HDL则无影响。电镜下观察,在OX-LDL的诱导下,SMC胞浆中的肌丝较对照组明显减少,而高氏器,内质网和线粒体则明显增多。上述结果说明,LDL、VLDL、OX-LDL、OX-VLDL和OX-HDL的致动脉粥样硬化作用可能与刺激SMC表型由“收缩”型向“合成”型转化有关。
On the basis of establishing in vitro culture methods of human aortic smooth muscle cells (SMCs), the effects of various natural and oxidized modified lipoproteins on the morphology and phenotype of human SMCs were observed. Under the action of oxidized low density lipoprotein (OX-LDL), oxidized high density lipoprotein (Ox-HDL) and oxidized low density lipoprotein (OX-VLDL), the morphological changes of SMC were as follows: , From spindle to irregular polygon, the process of protuberances was thin and long, and the growth pattern of “valley” and “peak” also changed. LDL and vLDL also affected the morphology of SMC but not as obvious as that of oxidized and modified lipoproteins; no effect. Under electron microscope, under the induction of OX-LDL, the myofilament in SMC cytoplasm was significantly decreased compared with the control group, while the Gallbladder, endoplasmic reticulum and mitochondria were significantly increased. The above results indicate that the atherogenic effects of LDL, VLDL, OX-LDL, OX-VLDL and OX-HDL may be related to stimulating SMC phenotypes from “contractile” to “synthetic”.
