【目的】观察青蒿琥酯作用于人急性白血病原代细胞后线粒体膜电位的变化。【方法】选取急性白血病患者的骨髓进行原代细胞培养,将终浓度为50、25、12.5μg/mL的青蒿琥酯分别作用于细胞,经线粒体膜电位荧光探针JC-1染色并固定后,在激光扫描共聚焦显微镜下观察细胞形态的变化,并计算红、绿荧光细胞数。【结果】不同浓度青蒿琥酯作用于人急性白血病原代细胞后,均可升高绿荧光细胞数,与空白对照组比较差异有显著性意义(P<0.05),并可不同程度降低红荧光细胞数(P>0.05),且呈一定的剂量依赖关系。【结论】青蒿琥酯抗急性白血病原代细胞的作用机制与降低其线粒体膜电位,进而使其胞内环境改变而使肿瘤细胞凋亡有关。
【Objective】 To observe the changes of mitochondrial membrane potential after artesunate acts on human acute leukemia primary cells. 【Methods】 Bone marrow of patients with acute leukemia were selected for primary cell culture. Artesunate at final concentrations of 50, 25 and 12.5 μg / mL were applied to cells and stained by mitochondrial membrane potential fluorescent probe JC-1 After that, the morphological changes of the cells were observed under a laser scanning confocal microscope and the number of red and green fluorescent cells was calculated. 【Results】 Artesunate at different concentrations increased the number of green fluorescent cells after treated with primary human acute leukemia cells, which was significantly different from that of the blank control group (P <0.05), and decreased to some extent Fluorescent cells (P> 0.05), and a certain dose-dependent relationship. 【Conclusion】 The mechanism of artesunate against acute leukemia primary cells is related to the reduction of mitochondrial membrane potential and thus to the change of intracellular environment and the apoptosis of tumor cells.
