The biosynthesis of gangliosides is catalyzed by a class of membrane-bound transferases. Catabolism (see Figure 1), ganglioside from the cell membrane into the lysosome transport process is not understood. Gangliosides progressively degrade from the hydrophilic end of the molecule under the action of lysosomal acid hydrolases. The recessive genetic defect of this hydrolase hinders substrate degradation and therefore deposits of undegraded material. Many sphingolipid hydrolases are key-specific hydrolases that degrade only a few related substrates (oligosaccharides, glycoproteins, and lipids).
