目的观察加入干扰素γ(IFN-γ)前后细胞自身分化抗原1(CDA1)在肺腺癌细胞株SPC-A1中的表达,探讨CDA1与p53、p21、周期素D1在SPC-A1中的表达的相关性。方法运用RT-PCR及蛋白印迹法检测加入IFN-γ前和1h及24h后CDA1、p53、p21、周期素D1mRNA及蛋白质的表达。结果肺腺癌细胞株SPC-A1中加入IFN-γ1h后,CDA1mRNA及蛋白的表达均增多,周期素D1mRNA的表达出现减少,而p53、p21的表达无明显改变;24h后,CDA1mRNA及蛋白的表达进一步增加,p53、p21的表达明显增加,而周期素D1mRNA及蛋白水平显著减少。结论CDA1是p53的上游基因,可能有抑癌基因的作用,对p35、p21及周期素D1的表达有调控作用。
Objective To investigate the expression of CDA1 in human lung adenocarcinoma cell line SPC-A1 before and after addition of interferon γ (IFN-γ) and to investigate the expression of CDA1, p53, p21 and cyclin D1 in SPC-A1 Relevance. Methods The expression of CDA1, p53, p21, cyclin D1 mRNA and protein were detected by RT-PCR and Western blotting before and 1h and 24h after adding IFN-γ. Results The expression of CDA1 mRNA and protein were increased in the lung cancer cell line SPC-A1 after IFN-γ1h treatment, while the expression of cyclin D1 mRNA was decreased but the expression of p53 and p21 was not changed. After 24 hours, the expression of CDA1 mRNA and protein Further increase, p53, p21 expression increased significantly, and cyclin D1 mRNA and protein levels were significantly reduced. Conclusion CDA1 is an upstream gene of p53 and may play a role in tumor suppressor gene regulating the expression of p35, p21 and cyclin D1.
