目的:研究壳聚糖对大鼠Ⅲ度烧伤创面脂质过氧化反应的影响及在创面愈合中的意义。方法:按Nagelschmidt法计算大鼠Ⅲ度烧伤后各时相点创面愈合率;用生化法测定各时相点创面组织蛋白、羟脯氨酸(HOP)、超氧化歧化酶(SOD)、丙二醛(MDA)含量。结果:4%壳聚糖组(w/v)各个时间点创面愈合率都明显高于自然愈合组(P<0.01);伤后7,14d,4%壳聚糖组织蛋白含量明显高于自然愈合组(P<0.01或P<0.05);2%壳聚糖组、4%壳聚糖组烧伤后7d HOP就达最高值,且其值比自然愈合组最高值大;伤后3,7,14d,2%壳聚糖组、4%壳聚糖组创面SOD活性比自然愈合组高(P<0.05);4%壳聚糖组创面MDA含量的升高比自然愈合组明显低(P<0.01或P<0.05)。结论:壳聚糖对大鼠Ⅲ度烧伤创面有促愈合作用,并呈剂量依赖性,其促愈合机制与使用壳聚糖后创面脂质过氧化反应程度减轻有关。
Objective: To study the effect of chitosan on lipid peroxidation in third-degree burn wounds of rats and its significance in wound healing. Methods: The rate of wound healing was calculated by Nagelschmidt method at each time point after burn injury of third degree. The histochemical, hydroxyproline (HOP), superoxide dismutase (SOD) Aldehyde (MDA) content. Results: The wound healing rate of 4% chitosan group (w / v) at each time point was significantly higher than that of natural healing group (P <0.01). At 7 and 14 days, the content of 4% chitosan tissue protein was significantly higher than that of natural healing group (P <0.01 or P <0.05). HOP in 2% chitosan group and 4% chitosan group reached the maximum at 7th day after burn, and its value was higher than that of natural healing group. After injury, 3,7 (P <0.05). The activity of SOD in wound surface of 4% chitosan group and 4% chitosan group was higher than that of natural healing group (P <0.05) <0.01 or P <0.05). CONCLUSION: Chitosan has a healing effect on third-degree burn wounds in a dose-dependent manner in rats, and its mechanism of promoting healing is related to the reduction of lipid peroxidation in wounds after chitosan treatment.
