目的建立苏州市区学龄前儿童全血锰(Mn)的参考范围。方法全血样经微波消解处理后,用等离子发射光谱法(ICP-AES)测定,参照NCCLS发布的临床实验室(C28-A2)的文件对儿童血锰含量数据进行统计处理,确定参考范围。结果本方法的批内CVw为1.6%~2.1%,批间CVb为2.0%~2.4%,偏倚CVbias为0.52%~0.68%,相对扩展不确定度为5.23%~6.52%,加标回收率在96.7%~98.1%之间。苏州市区学龄前儿童全血锰的参考范围为4.72~11.45μg/L(男:4.76~11.51μg/L,女:4.70~11.42μg/L)。锰元素参考范围在性别间的差异无统计学意义(P>0.05)。结论等离子发射光谱法测定全血锰,方法简单、可靠,建立的全血锰参考范围符合苏州市学龄前儿童的人群特点,可在临床推广应用。
Objective To establish a reference range of whole blood manganese (Mn) in preschool children in Suzhou. Methods Whole blood samples were processed by microwave digestion and analyzed by plasma emission spectrometry (ICP-AES) with reference to the C28-A2 file published by NCCLS to determine the reference range. Results The intra-assay CVw was 1.6% -2.1%, the inter-assay CVb was 2.0% -2.4%, the bias CVbias was 0.52% -0.68%, and the relative expanded uncertainty was 5.23% ~ 6.52% 96.7% ~ 98.1%. The reference range of whole blood manganese in preschool children in Suzhou city ranged from 4.72 to 11.45 μg / L (male: 4.76 to 11.51 μg / L, female: 4.70 to 11.42 μg / L). There was no significant difference in the reference range of manganese between the sexes (P> 0.05). Conclusion Determination of plasma whole blood manganese by plasma emission spectrometry is simple and reliable. The established reference range of whole blood manganese is in line with the characteristics of preschool children in Suzhou City, which can be popularized and applied clinically.
