[目的]研究母鼠孕前低水平铅暴露是否对子代骨骼钙代谢有影响。[方法]12只雌性昆明小鼠随机分为铅暴露组、对照组,每组6只。铅暴露组小鼠通过饮水暴露于醋酸铅(含铅元素10.0 mol/L)4周后怀孕。在仔鼠出生后的第21天和第60天,两组每窝随机选取3只仔鼠处死,取股骨、全血和血清,用石墨炉原子吸收法测定铅含量,用火焰原子吸收法测定钙含量,用放射免疫法测定血清总甲状旁腺激素(PTH)含量。[结果]在出生后第21天,与对照组仔鼠相比,铅暴露组仔鼠血钙水平[(1.75±0.24)mmol/L]、骨钙水平[(10.17±1.03)mmol/g]均明显降低,血铅水平[(0.31±0.03)μmol/L]、骨铅水平[(57.24±4.65)nmol/g]及血清总PTH水平[(3.40±0.74)μg/L]均明显升高(P<0.05)。到出生后第60天,与出生后第21天相比,铅暴露组仔鼠血钙水平[(2.11±0.17)mmol/L]明显上升(P<0.05),且与对照组仔鼠无明显差异(P>0.05);骨钙水平[(13.65±2.11)mmol/g]、血清总PTH水平[(5.91±0.85)μg/L]均明显上升(P<0.05),且与对照组仔鼠有明显差异(P<0.05);骨铅水平[(45.04±7.62)nmol/g]明显下降(P<0.05),但仍明显高于对照组仔鼠(P<0.05);血铅水平[(0.29±0.02)μmol/L]无明显改变,仍明显高于对照组仔鼠(P<0.05)。[结论]母鼠孕前低水平铅暴露影响导致了子代小鼠骨骼钙代谢。
[Objective] To study whether low level of lead exposure of female rats before pregnancy has an effect on calcium metabolism in offspring. [Methods] Twelve female Kunming mice were randomly divided into lead exposure group and control group, with 6 rats in each group. Lead exposure group mice were exposed to lead acetate (containing 10.0 mol / L lead) for 4 weeks after drinking. On day 21 and day 60 after the birth of the offspring, three litters were randomly sacrificed in each litter of two groups. Femurs, whole blood and serum were collected. The lead content was determined by graphite furnace atomic absorption spectrometry and determined by flame atomic absorption spectrometry Calcium content, radioimmunoassay for determination of serum total parathyroid hormone (PTH) content. [Result] Compared with the control group, the serum calcium level [(1.75 ± 0.24) mmol / L] and the bone calcium level [(10.17 ± 1.03) mmol / g] (0.31 ± 0.03) μmol / L], bone lead level [(57.24 ± 4.65) nmol / g] and serum total PTH level [(3.40 ± 0.74) μg / L] (P <0.05). After 60 days of birth, the level of serum calcium in the lead exposed group [(2.11 ± 0.17) mmol / L] significantly increased (P <0.05) compared with that in the control group (P <0.05). The levels of osteocalcin (13.65 ± 2.11) mmol / g and total PTH in serum [(5.91 ± 0.85) μg / L] (P <0.05). The levels of lead in bone [(45.04 ± 7.62) nmol / g] were significantly lower than those in control group (P <0.05) 0.29 ± 0.02) μmol / L], but still significantly higher than that of the control group (P <0.05). [Conclusion] The low level lead exposure of maternal pre-pregnancy led to calcium metabolism in offspring mice.
