目的探讨冬虫夏草多糖脂质体(CPL)抗肝纤维化的作用及机制。方法动物分为肝纤维化模型组、CPL治疗组和正常对照组,采用免疫组化和Northern杂交技术检测肝组织Ⅰ、Ⅲ型胶原蛋白及Ⅰ、Ⅲ型前胶原基因的表达。结果CPL治疗组大鼠肝脏中Ⅰ、Ⅲ型胶原蛋白较模型对照组明显减少(P值分别<0.01);CPL治疗组大鼠肝脏组织Ⅰ、Ⅲ型前胶原mRNA表达量明显下降,与模型组大鼠比较差异有显著性(P值分别<0.05)。结论冬虫夏草多糖脂质体能明显减少肝脏的胶原沉积,并能抑制Ⅰ、Ⅲ型前胶原基因的表达。
Objective To investigate the effect and mechanism of Cordyceps polysaccharide liposome (CPL) on hepatic fibrosis. Methods Animals were divided into hepatic fibrosis model group, CPL treatment group and normal control group. Immunohistochemistry and Northern blotting techniques were used to detect the expression of type I and III collagen proteins and type I and III procollagen genes in liver tissue. Results Compared with the model control group, the levels of collagen I and III in the liver of CPL-treated rats were significantly lower (P<0.01). The expression of type I and III procollagen mRNA in liver of CPL-treated rats was significantly decreased. There was a significant difference between the model group rats (P values <0.05). Conclusion Cordyceps sinensis liposome can significantly reduce collagen deposition in the liver and inhibit the expression of type I and III procollagen genes.
