观察自制抗组织因子途径抑制物单克隆抗体(McAb4F8)对血浆凝血酶原时间(PT)及活化的部分凝血活酶时间(APTT)的影响。结果发现:外源性的组织因子途径抑制物(TFPI)可使PT及APTT延长。McAb4F8则使稀释的PT缩短,且呈量效关系。低分子量肝素(LMWH)使PT及ATPP均明显延长,加入兔抗人抗凝血酶Ⅲ抗体则可以减弱LMWH的这种作用。McAb4F8还可使乏因子Ⅸ血浆稀释的PT明显缩短。提示组织因子途径在正常生理条件和血友病人的血凝过程中均起重要作用。
To observe the effect of McAb 4F8 on the prothrombin time (PT) and activated partial thromboplastin time (APTT). The results showed that extrinsic tissue factor pathway inhibitor (TFPI) prolonged PT and APTT. McAb4F8 shorten the diluted PT, and the dose-effect relationship. Low molecular weight heparin (LMWH) prolonged both PT and ATPP, and the addition of rabbit anti-human antithrombin III antibody attenuated this effect of LMWH. McAb4F8 also significantly shortened plasma-diluted PT of factor IX. Tissue factor pathways suggest that both normal physiological conditions and hemophilia hemophilia play an important role in the process.
