为了分析不同培养条件对美国山核桃花粉生活力的影响,以4个不同品种美国山核桃花粉为试验材料,研究了培养时间、培养温度、培养基p H、蔗糖、硼酸和PEG-4000浓度等对花粉离体萌发的影响。结果表明:不同培养时间对美国山核桃花粉的生活力有显著影响,适宜的培养时间为24 h;适宜的培养温度为25~30℃,温度过高或者过低均会抑制花粉萌发和花粉管生长;适宜的液体培养基p H为5.5,超过此范围均表现为抑制萌发。在一定浓度范围内,蔗糖、硼酸和PEG-4000对美国山核桃4个品种的花粉萌发和花粉管生长起促进作用,而超过阈值后起抑制作用;培养基组分中以含20%~25%的蔗糖、0.03%的硼酸和15%的PEG-4000为宜。
