目的本实验旨在探讨小窝蛋白在血管紧张素Ⅱ(AngⅡ)诱导的人系膜细胞收缩中的作用。方法人肾小球系膜细胞系用于全部实验。应用AngⅡ刺激经过或未经过甲基-β-环糊精(MβCD)预处理1小时的系膜细胞。采用时滞摄影技术检测细胞体积的变化。结果①应用1μmol/LAngⅡ刺激系膜细胞,细胞体积发生明显收缩(n=20,P<0.01)。②给予1μmol/LAngⅡ刺激经MβCD(10mmol/L)预处理1小时后的细胞,系膜细胞体积缩小不明显(n=20,P<0.01)。结论①血管紧张素Ⅱ能够诱导系膜细胞收缩。②Caveolae参与血管紧张素Ⅱ诱导的系膜细胞收缩。
Objective To investigate the role of caveolin in the contraction of human mesangial cells induced by angiotensin Ⅱ (AngⅡ). Methods Human mesangial cell lines were used for all experiments. Mesangial cells were pretreated with AngⅡ for 1 hour with or without methyl β-cyclodextrin (MβCD). Time-lapse photography was used to detect changes in cell volume. Results ① The mesangial cells were stimulated with 1μmol / L of AngⅡ, and the volume of cells was significantly contracted (n = 20, P <0.01). ② The volume of mesangial cells was not significantly reduced (P <0.01) after 1μmol / L of AngⅡ stimulated the cells pretreated with MβCD (10mmol / L) for 1 hour. Conclusion ① Angiotensin Ⅱ can induce mesangial cell contraction. ②Caveolae is involved in angiotensin Ⅱ-induced mesangial cell contraction.
