目的:探讨僵蚕天南星方对急性缺血性脑水肿大鼠水通道蛋白4(AQP4)表达的影响及其作用机制。方法:将大鼠随机分为3组:假手术组、模型组、僵蚕天南星方组,采用线栓法制备大鼠大脑中动脉缺血再灌注模型,观察缺血2 h再灌注24 h、48 h、72 h、96 h 4个不同时间点,采用Zausinger评分法观察大鼠神经功能改变,干湿重法测定脑组织含水量,HE染色观察脑组织病理学变化,免疫组织化学法观察大鼠大脑皮层AQP4的表达情况。结果:与模型组比较,僵蚕天南星方组神经功能评分较高(P<0.01),脑组织缺血性病理学损伤均明显改善,脑含水量降低(P<0.01),AQP4蛋白表达量减少(P<0.05)。结论:僵蚕天南星方能够改善脑缺血-再灌注损伤大鼠的神经功能缺损,可减轻脑缺血损伤引起的脑水肿,其机制可能与降低AQP4蛋白的表达有关。
Objective: To investigate the effect and mechanism of Anemonefina on the expression of aquaporin 4 (AQP4) in rats with acute ischemic cerebral edema. Methods: The rats were randomly divided into 3 groups: sham-operation group, model group and Najiantiannanxingfang group. Rat middle cerebral artery occlusion (MCAO) model was established by thread occlusion. 48 h, 72 h, 96 h four different time points, using Zausinger score method to observe the changes of nerve function in rats, wet and dry weight determination of brain water content, HE staining to observe the pathological changes of brain tissue, immunohistochemical observation of large Expression of AQP4 in rat cerebral cortex. Results: Compared with the model group, the scores of neurological function were significantly increased (P <0.01), the ischemic pathological changes of brain tissue were significantly improved, the brain water content was decreased (P <0.01), and the expression of AQP4 protein was decreased P <0.05). CONCLUSION: Zaoshen Tiannanxing can ameliorate the neurological deficit in rats with cerebral ischemia-reperfusion injury and relieve cerebral edema caused by cerebral ischemia injury, which may be related to the decrease of AQP4 protein expression.
