利用扫描电镜、描隧道显微镜及细胞荧光光度计,研究了有机锡化合物Et_2SnCl_2phen 对人胃癌细胞(BGC-823)膜表面结构、膜蛋白和核中 DNA 含量的影响。发现Et_2SnCl_2phen 不但能抑制细胞生长,而且能明显改变癌细胞膜表面结构。电镜下显示出癌细胞表面比较光滑和较少微绒毛。扫描隧道显微镜的图像显示出细胞表面不平和疏松分布。与对照组相比,经 Et_2SnCl_2phen 处理的细胞膜蛋白质和核中 DNA 的含量明显减少。
The effects of organotin Et_2SnCl_2phen on the surface structure, membrane protein and nuclear DNA content of human gastric cancer cell line BGC-823 were studied by scanning electron microscopy, scanning tunneling microscope and cytofluorimetry. Et_2SnCl_2phen was found not only inhibit cell growth, but also significantly change the surface structure of cancer cell membrane. Electron microscopy showed the surface of cancer cells is relatively smooth and less microvilli. Scanning tunneling microscope images show uneven and loose cell surface distribution. Compared with the control group, the content of DNA in the plasma membrane and nucleus treated with Et_2SnCl_2phen decreased significantly.
