目的:观察氟伐他汀对血管紧张素Ⅱ(AngⅡ)诱导的肾小管上皮细胞增殖的影响。方法:①不同浓度(10~(-9)、10~(-8)、10~(-7)、10~(-6)、10~(-5)mol/L)AngⅡ刺激体外培养的肾小管上皮细胞;②固定浓度AngⅡ(10~(-5)mol/L)刺激,不同浓度(10~(-9)、10~(-8)、10~(-7)、10~(-6)、10~(-5)mol/L)氟伐他汀干预;③在不同的时间点(24、48、72 h)用苯唑氮蓝法(MTT)检测细胞增殖。结果:①AngⅡ促进肾小管上皮细胞增殖,作用48 h增殖最明显,随着浓度增加,增殖作用增加,以10~(-5)mol/L效果显著;②氟伐他汀能够显著抑制AngⅡ的促增殖作用,同样作用48 h抑制作用最明显,随着浓度增加,抑制作用增加,10~(-5)mol/L效果显著。结论:氟伐他汀能抑制AngⅡ诱导的肾小管上皮细胞增殖作用,48 h作用最强,且具有一定的浓度依赖性。
Objective: To observe the effect of fluvastatin on the proliferation of renal tubular epithelial cells induced by angiotensin Ⅱ (AngⅡ). Conclusion: Fluvastatin can inhibit the proliferation of renal tubular epithelial cells induced by AngⅡ, and its effect is the strongest at 48 h, with a certain concentration dependence.
