目的:观察甲基化转移酶抑制剂zebularine对人胃癌SGC-7901细胞株抑癌基因p16表达的影响。方法:采用MTT法和划痕实验检测zebularine对人胃癌SGC-7901细胞株的抑制率及其细胞迁移率的影响,采用real-time PCR检测作用前后抑癌基因p16表达的变化。结果:⑴终浓度100μmol/L的zebularine作用24 h能显著抑制SGC-7901细胞株的增殖率和迁移率。MTT法对照组吸光度为0.50±0.01,实验组为0.45±0.02,两组间差异有统计学意义(P<0.05);对照组划痕愈合率为(47.45±10.05)%低于实验组的(88.23±5.45)%(P<0.05)。⑵zebularine作用24 h能显著增加抑癌基因p16mRNA的表达,实验组为5.21±0.67显著高于对照组的1.04±0.09(P<0.05)。结论:甲基化转移酶抑制剂zebularine可以有效抑制SGC-7901细胞株的增殖和迁移并提高抑癌基因p16mRNA的表达。
Objective: To investigate the effect of methylated transferase inhibitor zebularine on the expression of tumor suppressor gene p16 in human gastric cancer cell line SGC-7901. Methods: MTT assay and scratch assay were used to detect the effect of zebularine on the inhibition rate and cell migration rate of human gastric cancer cell line SGC-7901. Real-time PCR was used to detect the change of tumor suppressor gene p16. Results: (1) zebularine with a final concentration of 100 μmol / L for 24 h significantly inhibited the proliferation and migration of SGC-7901 cells. The MTT assay showed that the absorbance of the control group was 0.50 ± 0.01, the experimental group was 0.45 ± 0.02, the difference was statistically significant (P <0.05); the wound healing rate of the control group was (47.45 ± 10.05)%, lower than that of the experimental group 88.23 ± 5.45)% (P <0.05). (2) The effect of zebularine for 24 h significantly increased the expression of tumor suppressor gene p16 mRNA. The experimental group was 5.21 ± 0.67, which was significantly higher than that of the control group (1.04 ± 0.09, P <0.05). CONCLUSION: Methyltransferase inhibitor zebularine can effectively inhibit the proliferation and migration of SGC-7901 cell line and increase the expression of tumor suppressor gene p16mRNA.
