目的探讨二甲双胍对甲状腺乳头状癌IHH-4细胞增殖的影响及其机制。方法 MTT法分别检测不同浓度的二甲双胍(0、5、10mmol/L)和二甲双胍10mmol/L在不同时间点(0、24、48、72h)对乳头状癌IHH-4细胞增殖的影响。Western blot法检测不同浓度二甲双胍干预IHH-4细胞48h后单磷酸腺苷激活的蛋白激酶α亚基(AMPKα)及磷酸化AMPKα(p-AMPKα)蛋白表达。结果二甲双胍可呈浓度和时间依赖性抑制IHH-4细胞增殖(P<0.01)。随着二甲双胍干预浓度的增加,p-AMPKα/AMPKα表达量增加(P<0.01)。结论二甲双胍可通过激活AMPK通路抑制甲状腺乳头状癌IHH-4细胞的增殖活性。
Objective To investigate the effect of metformin on the proliferation of papillary thyroid carcinoma IHH-4 cells and its mechanism. Methods MTT assay was used to detect the effect of metformin (0,5,10 mmol / L) and metformin 10 mmol / L on the proliferation of papillary carcinoma IHH-4 cells at different time points (0, 24, 48 and 72 h). The expression of AMPKα and p-AMPKα in the IHH-4 cells treated with different concentrations of metformin 48h were detected by Western blot. Results Metformin inhibited the proliferation of IHH-4 cells in a concentration- and time-dependent manner (P <0.01). With the increase of metformin intervention, the expression of p-AMPKα / AMPKα increased (P <0.01). Conclusion Metformin can inhibit the proliferation of papillary thyroid carcinoma IHH-4 cells by activating AMPK pathway.
