目的:陈年菟丝子干种子常因进入次生休眠而影响发芽率,本实验探讨导致中药菟丝子休眠的主要原因,并寻求最佳处理方法。方法:单独处理与复配处理结合,运用正交设计,分析发芽实验结果。结果:陈年菟丝子干种子吸水能力良好;水浸液与甲醇提取物均有发芽抑制物,后者抑制作用较强;单独处理效果普遍不如复配处理。结论:种子抑制物的存在是种子休眠的主导因素,最佳的处理方法分别为:①浓硫酸处理2min后,用500mg·L-1的GA3溶液处理60min;②100mg.L-1的NaOH溶液处理20min后,用500mg·L-1的GA3溶液处理120min。与空白对照15.3%的发芽率相比,最高萌发率分别达到了43.3%,41.7%。
OBJECTIVE: The aged seeds of dried dodder often affect the germination rate due to the secondary dormancy. This experiment explores the main causes of the dormancy of Chinese dodder and seeks for the best treatment. Methods: Combining with single treatment and compounding treatment, the orthogonal design was used to analyze the germination experiment results. Results: The dried seeds of aged dried cotyledons had good water absorption capacity. Both aqueous extracts and methanolic extracts had the inhibitory effect of germination. The latter showed stronger inhibitory effect. The single treatment effect was generally inferior to that of compound treatment. Conclusion: The existence of seed inhibitor is the dominant factor of seed dormancy. The best treatment methods are as follows: ① After treated with concentrated sulfuric acid for 2min, treated with 500mg · L-1 GA3 solution for 60min; ②100mg.L-1 NaOH solution After 20min, treatment with 500mg · L-1 GA3 solution 120min. Compared with the blank control of 15.3% germination rate, the highest germination rate reached 43.3% and 41.7% respectively.
