目的:研究DKK-1(Dickkopf-1)在多发性骨髓瘤细胞中的表达水平,以及与骨髓瘤骨病发生的关系。方法:以DKK-1和骨髓瘤为检索词,检索1989-2009年中国知识资源总库CNKI系列数据库和PubMed数据库。纳入标准:1)DKK-1基因结构及生物学特征的研究;2)DKK-1与骨髓瘤发生关系研究。粗选有56篇关于DKK-1结构特点及其与骨髓瘤关系的研究文章,根据纳入标准,精选27篇文献。结果:DKK-1是Wnt/β-cate-nin信号通路的可溶性的抑制剂,目前已被认为在多发性骨髓瘤及骨髓瘤骨病的发生中起到一定作用。结论:中和DKK-1和(或)促进Wnt/β-catenin信号通路能够治疗骨病及抑制骨髓瘤细胞生长,提示DKK-1可能成为骨髓瘤骨病治疗的新靶点。
Objective: To investigate the expression of DKK-1 (Dickkopf-1) in multiple myeloma cells and the relationship with the occurrence of myeloma bone disease. Methods: Using DKK-1 and myeloma as the search terms, the CNKI series database and PubMed database of China Knowledge Base from 1989 to 2009 were searched. Inclusion criteria: 1) Study on the structure and biological characteristics of DKK-1 gene; 2) Study on the relationship between DKK-1 and myeloma. Crude selection of 56 articles on the structural characteristics of DKK-1 and its relationship with myeloma research articles, according to the inclusion criteria, selected 27 articles. Results: DKK-1 is a soluble inhibitor of Wnt / β-cate-nin signaling pathway and has been considered to play a role in the pathogenesis of multiple myeloma and myeloma. CONCLUSION: DKK-1 and / or promote Wnt / β-catenin signaling pathway can treat bone disease and inhibit the growth of myeloma cells, suggesting that DKK-1 may be a new therapeutic target for myeloma.
