目的:研究六味地黄丸含药血清对SD大鼠骨髓源性树突状细胞(bone marrow-derived dendritic cells,BMDC)增殖的影响。方法:制备六味地黄丸含药血清,观察含药血清作用下的细胞生长情况,进行细胞计数及细胞活力测定;用四甲基偶氮唑盐(MTT)法,检测不同浓度、不同时间六味地黄丸含药血清作用下的细胞增殖变化,寻找最佳六味地黄丸浓度和作用时间;ELISA法检测细胞上清中树突状细胞分泌细胞因子IL-12的浓度;混合淋巴细胞反应(MLR)检测其刺激T淋巴细胞活化和增殖能力。结果:与空白对照组比较,成熟型DC含药血清组显著促进DC细胞增殖,且这种增殖在含药浓度15%,作用时间48 h时最为显著。其IL-12分泌量显著提高(P<0.05)。六味地黄丸含药血清培养的成熟型DC对异基因T淋巴细胞具有增殖刺激作用,且随着细胞数量的增加作用增强(P<0.05),呈量效关系。结论:六味地黄丸含药血清具有诱导大鼠骨髓源性树突状细胞增殖的作用。
Objective: To study the effect of Liuwei Dihuang Pills containing serum on the proliferation of bone marrow-derived dendritic cells (BMDC) in SD rats. Methods: The drug-containing serum of Liuweidihuangwan was prepared and the growth of cells under the action of drug-containing serum was observed. The cell count and cell viability were measured. MTT assay was used to detect the inhibitory effect of Liuweidihuang Pill The concentration and the action time of the Liuweidihuang Pill under the action of the drug-containing serum were observed; the concentration of IL-12 secreted by dendritic cells in the supernatant of the cells was detected by ELISA; the mixed lymphocyte reaction (MLR) It stimulates T lymphocyte activation and proliferation. Results: Compared with the blank control group, the mature DC-containing serum group significantly promoted the proliferation of DCs, and the proliferation was most significant at the concentration of 15% and the action time of 48 h. The IL-12 secretion was significantly increased (P <0.05). The mature DCs cultured with Liuweidihuang Pill serum had proliferation and stimulation effects on allogeneic T lymphocytes, and increased with the increase of cell number (P <0.05). Conclusion: Liuwei Dihuang pill containing serum can induce the proliferation of rat bone marrow-derived dendritic cells.
