目的:建立老鹰茶药材中黄酮类成分的含量测定方法。方法:运用加压溶剂提取技术和HPLC-DAD对4批不同产地的老鹰茶药材进行分析。色谱条件:Agilent ZORBAXSB-C18(250mm×4.6mm,5μm)色谱柱,以水-甲醇为流动相梯度洗脱,流速1.0mL.min-1,检测波长350nm,柱温30℃。结果:7个黄酮(金丝桃苷、异槲皮苷、山柰酚-3-O-β-D-半乳糖苷、槲皮苷、紫云英苷、槲皮素和山柰酚)在一定浓度范围内线性关系良好(r>0.999);具有较好的精密度(日内精密度与日间精密度分别为3.9%和4.6%);回收率在91.6%~105.6%之间。不同产地老鹰茶药材黄酮类成分含量明显不同。结论:该方法可用于老鹰茶中黄酮类成分的定量分析。
Objective: To establish a method for determination of flavonoids in eagle tea medicinal materials. Methods: Four batches of eagles tea from different areas were analyzed by pressurized solvent extraction and HPLC-DAD. Chromatographic conditions: Agilent ZORBAXSB-C18 (250mm × 4.6mm, 5μm) column with water-methanol as the mobile phase gradient elution, flow rate 1.0mL.min-1, detection wavelength 350nm, column temperature 30 ℃. RESULTS: Seven flavones (hyperin, isoquercetin, kaempferol-3-O-β-D-galactoside, quercitrin, albin, quercetin and kaempferol) The linearity was good in a certain concentration range (r> 0.999). The precision was good (the intra-day and inter-day precision were 3.9% and 4.6%, respectively). The recoveries ranged from 91.6% to 105.6%. Different origin Hawthorn herbs flavonoids content was significantly different. Conclusion: This method can be used for quantitative analysis of flavonoids in eagle tea.
