目的 :研究白细胞介素 - 2 (IL - 2 )和白细胞介素 - 12 (IL - 12 )活化骨髓的抗肿瘤效应与重建造血功能 ;探讨活化骨髓的性质和杀伤肿瘤细胞的机制。方法 :测定活化骨髓杀伤肿瘤细胞的活性 ;检测活化骨髓的细胞表面分化抗原 ,特别是CD34+/CD38-细胞群的表达。结果 :活化骨髓与L12 10 细胞、新鲜白血病细胞共同培养后显示有杀伤肿瘤细胞作用 ,以培养 3～ 4天时最强 ;活化骨髓细胞的CD3、CD8、CD16、CD5 6分化抗原表达与新鲜骨髓相比升高 ,有显著性差异 (P <0 0 1)。L12 10 细胞、新鲜白血病细胞与活化骨髓共同培养后分化抗原CD71、CD95表达下降 ,超二倍体、亚二倍体等异倍体减少 ,与对照组相比有显著性差异 (P <0 0 5 ) ;活化骨髓中CD34+/CD38-细胞分化抗原表达与新鲜骨髓相比没有显著性差异 (P >0 0 5 )。IL - 12活化骨髓效靶比 (E∶T)稀释一倍后杀伤白血病细胞活性仍然比IL - 2活化骨髓明显 ;IL - 2联合IL - 12诱导后活化骨髓抗肿瘤活性进一步增强。结论 :活化骨髓具有抗肿瘤效应 ,其中有细胞毒T细胞、自然杀伤细胞的作用 ,也有凋亡机制参与 ;活化骨髓还保留有重建造血的功能 ;IL - 12活化骨髓比IL - 2活化骨髓的活性高 ;且IL - 2联合IL - 12诱导后活化骨髓抗肿瘤活性进一步增强
Objective: To study the antitumor effects of interleukin - 2 (IL - 2) and interleukin - 12 (IL - 12) activated bone marrow and to reconstruct the hematopoietic function. To investigate the properties of activated bone marrow and its mechanism of killing tumor cells. Methods: The activity of activated bone marrow-killed tumor cells was measured. The expression of cell surface differentiation antigen (CD34 + / CD38-), especially CD34 + / CD38- cells in activated bone marrow was detected. Results: The activated myeloid cells and L12 10 cells co-cultured with fresh leukemia cells showed the cytotoxic effect on tumor cells, which was the strongest in 3 to 4 days. The expression of CD3, CD8, CD16 and CD5 6 in activated myeloid cells was positively correlated with fresh myeloid phase There was a significant difference between the two groups (P <0.01). L12 10 cells, fresh leukemia cells and activated bone marrow differentiated antigen CD71, CD95 decreased expression of diploid, sub-diploid aneuploid decreased compared with the control group were significantly different (P <0 0 5). There was no significant difference in the expression of CD34 + / CD38- cell differentiation antigen in activated bone marrow compared with fresh bone marrow (P> 0.05). The activity of leukemic cells was still significantly lower than that of IL - 2 - activated bone marrow when the ratio of activated bone marrow targets (E: T) was doubled with IL - 12. The anti - tumor activity of activated bone marrow activated by IL - 2 combined with IL - 12 was further enhanced. CONCLUSION: Activated bone marrow has anti - tumor effect, including cytotoxic T cells and natural killer cells, and also has apoptotic mechanisms. Activated bone marrow also retains the function of rebuilding hematopoiesis.
