目的对乌拉立肽粗品纯化并制备符合纯度要求的乌拉立肽纯品。方法采用C18(H)ODS柱(30mm×250mm,10μm);流动相A为0.1%甲酸-水溶液,流动相B为100%乙腈;检测波长215nm;体积流量30mL/min;进样量60mg。结果 RP-HPLC一步纯化后得到乌拉立肽质量分数达到98%。通过多次样品收集和冷冻干燥,制备了高纯度的乌拉立肽。结论建立了乌拉立肽的纯化工艺,制备出符合要求的乌拉立肽纯品。
OBJECTIVE To purify the crude product of uralizarin and prepare pure uralizarin which meets the purity requirements. Methods The mobile phase A was 0.1% formic acid-water, the mobile phase B was 100% acetonitrile, the detection wavelength was 215 nm, the volume flow rate was 30 mL / min and the injection volume was 60 mg. Results One-step purification of RP-HPLC resulted in the mass fraction of uralizarin up to 98%. Highly purified ularitide was prepared by multiple sample collection and freeze-drying. Conclusion The purification process of ularitin was established, and the pure uralizide was prepared to meet the requirements.
