探讨60Coγ射线照射对成纤维细胞增生和Ⅰ型胶原合成的作用机制。方法将人胚肺成纤维细胞(HELF)以不同剂量照射和用血管紧张素Ⅱ(AⅡ)刺激,测定培养液中丙二醛含量,检测细胞增生程度,用免疫细胞化学显示AⅡ和Ⅰ型胶原的合成。结果1~5Gy照射能刺激细胞增生和Ⅰ型胶原合成,同时促进胞内AⅡ合成;外源性AⅡ能促进细胞增生及胶原合成。结论HELF经60Co照射能过多合成AⅡ,其可能与照射后细胞增生有关
To investigate the mechanism of 60Coγ-ray irradiation on fibroblast proliferation and type Ⅰ collagen synthesis. Methods Human embryonic lung fibroblasts (HELF) were irradiated with different dose and stimulated with angiotensin Ⅱ (AⅡ). The content of malondialdehyde in the culture medium was measured to detect the degree of cell proliferation. Immunocytochemistry was used to detect the expression of AⅡ and type Ⅰ collagen Synthesis. Results 1 ~ 5 Gy irradiation can stimulate cell proliferation and collagen Ⅰ synthesis, while promoting intracellular A Ⅱ synthesis; exogenous A Ⅱ can promote cell proliferation and collagen synthesis. Conclusion HELF can synthesize AⅡ by 60Co irradiation, which may be related to cell proliferation after irradiation
